Long read의 필요성이 강조되고 있는 또 하나의 과제, Human gene duplication 연구에 대한 논문을 소개해드립니다. 

중복 유전자가(duplicate gene) 진화 적응적 측면에서 중요하다고 알려졌음에도 불구하고 부분복제 (segmental duplication) 구역에서의 정확한 주석이 부족한 경우가 많습니다. 연구팀은 이같은 중복 구간을 이해하기 위하여 Long read sequencing을 이용하였습니다. 

PacBio의 long read RNA seq을 이용한 full length transcript sequencing은 반복 구간에서도 충분한 숫자의 paralogous sequence variants를 확인할 수 있고, 발현 빈도가 낮은 구간에서도 whole transcriptome RNA-Seq에서 확인되지 않는 duplicate gene들을 분석할 수 있습니다. 

본 논문에서는 IsoSeq과 Hybridization 기술을 이용하여 사람 특이적 분절 유전자 (Human specific segmental genes)에 의해 전사되는 이성체(isoforms)의 서열을 분석하였고, 그 결과 수십개의 새로운 이성체를 발견하였습니다.
