인간 유전체에서 아직 해석이 미진한 “Dark" 영역에는 임상적으로 중요한 유전자가 많이 있습니다. 이러한 영역은 short read sequencing
data 또는
mapping의 어려움 등으로 coverage가 부족하여 아직 미스터리로 남아 있습니다.
이 영역에서 다양한 반복 영역 또는 한쪽으로 치우쳐진 염기의 구성으로 인해 NGS sequencing
yield가 낮아질 수 있고 단편 복제(segmental duplication)로 인한 정확하지 않은 mapping이 이루어질 수 있습니다.
PacBio의 최근 개선된 sequencing chemistry와 기기 성능으로 새로운 PacBio 데이터 유형, 매우 정확한 long read(HiFi read)를 생성합니다. 이 read들은 short read에 견줄 수 있는 정확도를 가지면서 read 길이는 50-100배 더 깁니다. 따라서 PacBio HiFi long read
