Next-Generation Cytogenetics: international, multi-center study of chromosomal aberrations in constitutional diseases and leukemia with Bionano Genome imaging
구조변이 (SV)는 인간게놈에서 다수의 질병 및 암에 연관된 유전자 변이의 중요한 원인입니다. SV는 인간 게놈의 repeat –rich 영역에서 많이 분포하고 있고 통상적인 short-read sequencing으로 검출되지 않은 채 남아있는 구간이 있습니다. Bionano Genomics의 고해상도 광학 매핑을 적용하여 포괄적으로 구조변이를 식별한 사례 및 아직 발견이 되지 않은 구조 변이를 어떻게 분석할 수 있는지를 소개합니다.
a) 52개의백혈병 샘플에서 체세포 돌연변이 감지할 수 있는 deep-genome coverage (400x) : 52 개의 백혈병 샘플에서 체세포 구조변이 분석한 결과는 기존에 수행이 되어온 진단분석과 일치
b) 매우 높은 해상도 (≥500bp) 및 시퀀싱 바이어스가 없기 때문에 희귀 질환에서 찾기 힘든 구조변이 탐지 : tumor-predisposition syndrome에서 retrotransposon insertion 확인
백혈병의 진단 표준 분석과의 완전한 일치는 고전적인 세포 유전 학적 시험의 대체 가능성을 보여주고 있습니다.
